免疫原

10个氨基酸的肽,对应于与卵清蛋白偶联的小鼠脑源性神经营养因子(BDNF)的氨基末端。序列为H2N-HSDPARRGEL-COOH。

应用

免疫组化:1-10 μg/mL(请参阅建议的方案)。

免疫印迹:1-10 μg/mL。BDNF在标准缓冲液中不易提取。切开的组织应在10倍体积的100mM磷酸盐缓冲液中匀浆,该缓冲液包含1mM EDTA,2M盐酸胍(pH7.2)和三种蛋白酶抑制剂,10mM N-乙基马来酰亚胺,0.36 mM胃抑素和1mM PMSF。然后对匀浆进行超声处理,并在4°C下以46K x g离心30分钟。 BDNF迁移为18 kDa,同型二聚体约为27-30 kDa。
