应用

磷酸酶抑制剂混合物套装III已用于：
  • 在三倍洗涤剂缓冲液中裂解培养细胞，提取总蛋白用于蛋白质印迹分析[1]
  • 作为裂解缓冲液的组成部分、裂解组织样品或培养的大鼠心室心肌细胞（VCM），用于心肌线粒体蛋白的蛋白质印迹分析[2]
  • 作为裂解缓冲液的组成部分，裂解感染Adv-COX7RP和Adv-shRNA COX7RP的培养大鼠VCM，用于免疫沉淀莱诺二氏受体2型（RyR2）[2]
  • 作为冰冻RIPA缓冲液中的一种成分，匀质化/裂解视网膜组织，用于蛋白提取和蛋白质印迹分析[3]

生化/生理作用

蛋白磷酸化和去磷酸化在调节细胞分裂、增殖和凋亡等多种细胞功能的信号转导过程中起重要作用。蛋白激酶将磷酸基团转移到特定的丝氨酸、苏氨酸或酪氨酸残基上，导致蛋白质的构象改变并影响其与配体的相互作用。蛋白质磷酸酶通过去除磷酸基团并将蛋白质恢复到其原始的去磷酸化状态来逆转这一过程。当从细胞和组织裂解物中提取磷酸化蛋白时，必须保持蛋白质的磷酸化状态。
