General description
使用人胎盘胃蛋白酶提取物，通过连续盐沉淀、乙醇沉淀和的 DEAE 色谱法纯化的人 I 型胶原蛋白。胶原蛋白是结缔组织的主要结构蛋白，存在于皮肤、肌腱、软骨、韧带、骨骼、眼球的白色部分（巩膜），以及细胞与组织之间的空间（称为细胞外基质）。它赋予结缔组织的结构和强度。I 型胶原蛋白α1 （COL1A1）基因位于人染色体17q21.33。I 型胶原蛋白在细胞中以前胶原蛋白的形成生成。该蛋白质由两条前体 α1（I）蛋白链与一条前体 α2（I）胶原蛋白前体链结合而成，形成三链绳状结构。在细胞中时，酶通过添加三链形成稳定分子并在链之间建立连接（交联）所必需的化学基团来修饰蛋白质中的某些氨基酸（赖氨酸和脯氨酸）。其他酶会在蛋白质中添加糖。现已完成，三链 I 型胶原蛋白前体分子离开细胞，并通过酶将两端的小片段剪断。原胶蛋白前体分子在细胞外部排列成细长的原纤维。原纤维并排在一起形成胶原纤维。原纤维中分子之间的交联使得蛋白质结构非常稳定，这有助于胶原蛋白的组织增强功能。
Application
人I型胶原蛋白已用于：
- 活化T细胞2B4核因子（NFAT)）–GFP报告基因细胞检测中的对照品，在该检测中其与报告基因细胞的相互作用可以评估活化T细胞核因子（NFAT）启动子驱动的GFP表达
- 在动态结合测定中包被玻璃载玻片，以构建特异性结合的底物并研究流动通道中的血小板和结合物
- 作为肺组织样品组织学分析中的蛋白质标准品，为细胞外基质（ECM）成分的组成和标准提供参考。
Biochem/physiol Actions
I型胶原蛋白α1（COL1A1）基因的突变与I–IV型成骨不全症、VIIA 型 Ehlers-Danlos 综合征、经典型 Ehlers-Danlos 综合征、Caffey 病和特发性骨质疏松症有关。
Physical form
纯化的蛋白质。 含有 0.5 M 醋酸，pH 2.5 的液体。应用时可以在 PBS 中稀释。
