白血病抑制因子(LIF)是一种多效性糖蛋白,最初被发现在诱导分化成巨噬细胞时,可抑制小鼠骨髓白血病细胞系M1的增殖。LIF的其他活性后来被陆续发现,人们可能了解其各种同义名,包括DIF、D因子、DIA、DRF、CNDF、HILDA、HSF-III和MLPLI。人LIF通过包含190kDa LIF结合α-链(130kDa,小鼠)和130kDa信号转导β-链(gp130)的受体发挥其作用,其与CNTF、OSM、L-6和IL-11公用同一受体。因此,它是细胞因子受体超家族gp130家族的成员。LIF受体已在几种细胞中得到鉴定,包括单核细胞、肝脏、胎盘和胚胎干细胞。天然LIF是高度糖基化的,研究显示根据来源不同,其分子量为32kDa至62kDa,但是没有糖基化似乎不影响其生物活性。LIF由单个基因编码,在人或小鼠中其分泌为含有180个氨基酸的单链糖蛋白,并具有保守的二硫键。人和小鼠LIF共享78%的序列同源性。人LIF可激活小鼠细胞,但小鼠LIF不能激活人体细胞。
