一般描述
紧密连接蛋白 ZO-1（UniProt Q07157；也称为紧密连接蛋白 1、Zona occludens 蛋白 1、闭锁小带蛋白 1）在人类中由 TJP1 基因（也称为 ZO1）（基因 ID 7082）编码。紧密连接（TJ） 构成了质膜的顶端和基底外侧区域之间的屏障。该屏障的组装和渗透性取决于闭锁小带（ZO） 膜相关鸟苷酸激酶（MAGUK） 蛋白 ZO-1、ZO-2 和 ZO-3。 ZO-1 是一种 210-225 kDa 的蛋白质，存在于上皮和内皮 TJ 的膜下结构域。ZO-1 在其 N 末端包含 3 个 PDZ 结构域（PDZ1/aa23-109；PDZ2/aa181-261；PDZ3/aa422-502），然后是一个 SH3 结构域（aa519-580），一个 GUK/GK 同源结构域（aa632-782）、酸性结构域（aa817-894）、α 剪接结构域（aa921-1000） 和 C 端富含脯氨酸/PR 结构域。ZO-1 是一种磷蛋白，是丝氨酸/苏氨酸激酶 ZAK 和 PKC 的已知底物。 MAPK 信号通路调节 ZO-1 的酪氨酸磷酸化，据报道，在 Ras 转化的上皮细胞中抑制 MEK1 会导致 ZO-1 和闭锁蛋白的酪氨酸磷酸化。 ZO-1 通过其 PDZ 结构域与 claudins、JAM、ZO-2 和 ZO-3 相互作用，而其 GK 模块介导与闭锁蛋白的相互作用。ZO-1 还通过其羧基末端结合肌动蛋白细胞骨架和肌动蛋白结合蛋白 4.1。此外，ZO-1 与 TJ 处的 AF-6 和 cingulin 以及粘附连接蛋白 α-连环蛋白以及间隙连接蛋白连接蛋白 43 和 45 相关联。
