对于胰蛋白酶消化肽，胰蛋白酶:肽的使用比例约为1:100至1:20。本品的典型用途是使贴壁细胞从培养表面脱落。 使细胞解离所需的胰蛋白酶浓度主要取决于细胞类型和培养物代数。 Trypsins也被用于细胞培养过程中的细胞再悬浮，在蛋白质组学研究中用于蛋白质的消化，以及在各种凝胶消化中。 其他的应用包括通过基于膜的技术评估结晶，以及在一项研究中确定蛋白质折叠速率和产量可以被动力学陷阱的存在所限制。

Biochem/physiol Actions

胰蛋白酶可切割赖氨酸和精氨酸残基的C末端肽段。 如果酸性残基位于切割位点的任意一侧，则该反应的水解速度减慢，并且如果脯氨酸残基位于切割位点的羧基侧，则水解停止。 胰蛋白酶活性的最佳pH条件为7-9。 胰蛋白酶还可以切割氨基酸合成衍生物的酯和酰胺键。 将EDTA添加到胰蛋白酶溶液中作为螯合剂，用于中和钙离子和镁离子以防止其抑制胰蛋白酶活性。 去除这些离子会增加酶活性。

丝氨酸蛋白酶抑制剂（包括DFP、TLCK、APMSF、AEBSEF和抑肽酶等）会抑制胰蛋白酶。

Disclaimer

1 mM HCl 溶液等分试样可在-20°C 条件下稳定储存 1 年。Ca2+的存在也会减弱胰蛋白酶的自溶，维持其在溶液中的稳定性。胰蛋白酶在 2.0 M 尿素、2.0 M 盐酸胍或 0.1% (w/v) SDS 中也保留大部分活性。
