一般描述

3-异丁基-1-甲基黄嘌呤/IBMX是cAMP和cGMP磷酸二酯酶的非选择性、非特异性抑制剂。IBMX可诱导黑色素生成,并可以用作黑色素生成研究的阳性对照。 在卵母细胞研究中,IBMX有助于维持前期I卵母细胞的生发泡期(GV)阻滞。

应用

3-异丁基-1-甲基黄嘌呤已用于:
  • 分化间充质干细胞 (MSC)
  • 用作诱导脂肪形成的培养基补充剂
  • 用作Krebs-Ringer缓冲液和HEPES (KRBH)溶液的补充剂,诱导葡萄糖刺激的胰岛素分泌(GSIS)
  • 鉴定脂肪来源的间充质干细胞
  • 环磷酸腺苷(cAMP)测定
  • 磷酸二酯酶(PDE)抑制

生化/生理作用

由IBMX对磷酸二酯酶的抑制而引起的cAMP水平的增加可激活PKA,而引起增殖减少、分化增加并诱导细胞凋亡。IBMX可抑制苯肾上腺素诱导的气道粘膜神经内分泌上皮细胞5-羟色胺释放。(IC50:1.3 μM)。IBMX还可作为一种腺苷受体拮抗剂。其已表现出对神经肌肉接头、GH3细胞和血管平滑肌细胞中离子通道的抑制作用。IBMX可诱导感觉神经元释放储存在细胞内的钙。
